I am using on my 3 column layout, a 300x250 square inside the content, a 160x600 block on the left sidebar, and a 250x250 block on the left sidebar. What do you think?

If you have a site with blocks of content (like a blog), a 300 x 250 between posts to break up the content can be a great placement for publishers, in my experience. It's especially great if that placement is above the fold.

BTW, did you guys see the Browser Size tool that was announced on the Google blog (and the Google Code blog)? It seems like a great resource for webmasters if you're curious to see exactly what content (and ad units) on your pages are above the fold for different users.

Well, kinda. It doesn't work with centered sites, and there are other issues. I look forward to version 2.
